Day 1 - Tuesday 12th July The Fundamental Techniques
Talks & techniques
The basics: Introduction to the chicken facility, the husbandry behind fertile egg production for science, 3Rs, Home Office Regulations, good egg incubation techniques, windowing and staging
Introduction to the fluorescent chicken lines: ‘Roslin Green’ eGFP, ‘Flamingo’ tdTomato, ‘Chameleon’ minibow, Membrane GFP
Ex ovo culture: 'New' Culture, EZY Culture, Shell-less late-stage Culture, Cultured organ slices
Application of substances to embryos: Beads, teratogens, drugs and inhibitors, including TAT-Cre to ‘Chameleon’ chick embryos

Day 2 - Wednesday 13th July New and Eggciting Times
Talks & techniques
Genome engineering: Generation and use of sterile surrogate hosts, Primordial Germ Cell (PGC) isolation, gene editing and culture
In ovo transfection: Overexpression, knock-out/down techniques, electroporation, siRNA
Grafting tissues: Cutting, pasting and chimera generation, the production of specialised tools for tissue grafting
Public Engagement and Teaching with the chicken embryo (optional)

Day 3 - 14th Thursday July Free Form Lab Work, Genomics and Gene Expression
The morning will be reserved for ‘free-form experiments’ for participants to try out techniques and generate preliminary data.

Chicken genomics: The state of play of the avian genome, genetic networks, functional annotation of the embryonic chick genome
Gene Expression Techniques: RNA in situ hybridisation-HCR, RNAScope

Evening
Open floor discussion & flash talks. Sandpit discussions to establish the forward momentum of the field, what are the needs in terms of resources, technology development, and joint funding bids. Food and drinks provided.

Follow up on practical work and discussions in the morning.
